Abstract

The molecular marker is a useful tool for assessing genetic variations and resolving cultivar identities. The objective of this study was to evaluate the genetic diversity of 18 germplasms tuberose (Polianthes tuberosa L.) using RAPD. In RAPD Analysis eight primers generated a total of 70 polymorphic bands. The number of amplification products produced by each primer varied from 2 to ?.Genetic diversity of 18 germplasm estimated by polymorphic information content (PIC) value ranged from 0.86 to 0.99. The cluster dendrogram, constructed on the basis at RAPD data showed similarity values from 0.52 to 0.99. Dandrogram generated using RAPD data showe..d two major clusters. Cluster II was the largest and included 14 germplasms while clusters I was comprised of four germplasms. Dendrogram revealed that the variety Sikkim selection was distantly located to Hyderabad single with similarity coefficient of 0.44. In current studies RAPD primers namely OPW-13, OPN-20 and OPA-10 generated higher level of polymorphism and higher level of PIC values. Hence these RAPD primers can be efficiently used for the estimation of molecular characterization in Tuberose.
