Isolation and Identificaiton of Microorganism from Faecal Samples in Poultry

Abstract
The purpose of this study was to isolate and identify microorganisms from chicken
faeces. The complete research project was carried out between February and April in
the laboratory of Division of Biological Standardisation, Indian Veterinary Research
Institute. lzntnagar, Bareilly. 30 faeces samples were used in the investigation. The
entire testing process was carried out in sterile conditions. By examining colony shape,
gram's staining and biochemical tests. isolates and their identities were determined. E.
coli colonies on MacConkey agar- showed smooth pinkish colony. Salmonella sp.
colonies on Hektoen Enteric agar were smooth black centred colonies. Confmnation of
E. coli isolates were done by biochemical test. The E. coli isolates showed positive test
for Indole, Catalase and MR while negative test for VP, Oxidase and Citrate. The
Salmonella sp showed negative for Indole, VP, oxidase and positive for MR, citrate,
catalase. A total of 24 E. coli and 6 Salmonella sp were isolated from the 30 faecal
samples. The 24 E. coli isolates were found positive for fimC (496 bp) and 16S rRNA
(585 bp) genes.
