Comparative Analysis of Genetic Diversity Among Finger Millet Genotypes for Salt Tolerance Using Molecular Markers

Abstract

Present study was undertaken to screen the 25 Finger-millet (Eleusine coracana) genotypes for salt Tolerance and assessment of genetic diversity with the help of 9 Rice-SSR primers and 10 CYP450 molecular markers. Tlu·ee different treatments of Salt (NaCI) i.e. 50rnM, 100mM and 150mM were used to create the osmotic stress, which reveal that out of these PRM 8113 shows maximum germination, PRM 81 04 shows maximum root length, PES 400 shows maximum shoot length, PRM 8104 shows maximum total length, PRM 8124 shows maximum root fresh weight, PRM 8112 shows maximum shoot fresh weight and PRM I shows maximum total fresh weight, PES 400 shows maximum root dry weight, PRM 6124 shows maximum shoot dry weight, PRM 6124 shows maximum total dry weight and PRM 8104 shows maximum seedling vigor index. And in genetic diversity analysis two types of molecular markers, SSR and CYP450 were assayed. In SSR analysis PIC values varies from 0 (RM 12) to 0.9984 (SSR 4) with average of 0.4992 and resolving power varies from 0.32 (RM 138) to 2.4 (RM 182) with average of 1.36. In CYP450 analysis PIC values varies from 0.8978 (Cyt15, 16) to 0.9984 (Cyt07, 08) with average of0.9481 (Fig 4.27) and resolving power varies from 0.16 (Cyt07, 08 and Cyt09, 010) to 3.12 (Cyt 05, 06) with average of 1.64. The two molecular marker systems together provided wider genome coverage and were found to be better indicator of the genetic relationships. No association was found between clustering of genotypes on the basis of the molecular marker analysis and on the basis of morphological parameters related to salt stress
