Abstract
Genetic variability among 7 accessions of fenugreek (Trigonella foenum-graecum) were subjected to
random amplified polymorphic DNA (RAPD) analysis using polymerase chain reaction with 30 arbitrary
decamer oligonucleotide primers. Fenugreek genotypes included in present study were from seven
regions of western U. P .. A total of 65 marker bands were observed out of which, 41 were polymorphic.
Out of 30 primers, 11 primers generated 100% generated and give scorable and reproducible results,
hence considered for genetic diversity analysis. Remaining primers generated 80%,75%,60%,50% and
0% polymorphic bands. The genetic diversity was calculated for each primer, which varied from 0.133 to
0.898, with a mean value of 0.352. Resolving power ranges from 1.14-9.14 in case of Trigonella foenumgraecum.
Genetic similarity estimates (Jaccard's coefficient) based on RAPD banding pattern were used
for cluster analysis to present genetic relationship in the form of dendrogram. The unweighed pair group
for arithmetic average analysis (UPGMA) was used for cluster analysis. The range of similarity was
found between 0.459(v4,v7) to 0.796(v4,v6). The maximum similarity was found between accessions
collected from Bijnor and Baghpat and minimum similarity was found between accessions collected from
Bijnor and Ghaziabad.
