Abstract
Studies were conducted to evaluate the genetic diversity of twenty genotypes of Aloe Vera
using the SSR and RAPD markers. RAPD analysis with ten primers generated a total of
I36 polymorphic bands. The number of amplification products produced by each primer
varied from 1 to 3.Genetic diversity of twenty genotypes in RAPD analysis as estimated
by polymorphic information content (PIC) value ranged from 0.52 to 0.96. The cluster
dendrogram of RAPD showed similarity values from 0.65 to 0.92. Dandrogram generated
using RAPD data showed two major clusters. Cluster I consist of two genotypes however,
cluster II included eighteen genotypes. Dendrogram revealed that Rishekesh Aloe-I was
distinctly related with home Aloe at a similarity coefficient 0.54. SSR analysis generated a
total of 3 7 bands with twenty primers. The number of polymorphic allele ranged from I to
4· with different primers. Out of twenty primers eight primers showed I 00%
polymorphism wllile, no result was obtained with eight SSR primers. Genetic diversity
using SSR data as estimated by PIC values ranged from 0.65 to 0.99. The cluster
dendrogram of SSR revealed 2 major clusters. Cluster II was the largest and included 18
genotypes while clusters I comprising of two genotypes namely Pant-2 and pant-3.
Dendrogram generated using SSR data showed that Rishekesh Aloe-3, Pant-4 and
Sahjahanpur Aloe-3 were genetically similar. Dendrogran1 of combined analysis revealed
2 major clusters. Cluster II was largest and included I6 genotypes while, clusters I
comprising of four genotypes. It was observed that RAPD markers can be successfully
used to study the genetic variation in Aloe vera as all the RAPD primers used in current
studies showed 100% polymorphism. It was found that SSR primers namely
RM1150,MCPCIR-l,MCPCIR-2 and MCPCIR-3 can be successfully used to study the
genetic variation in Aloe Vera. The information generated from current studies can be
used in future breeding programs of aloe vera improvement for various traits.
